[摘要]目的 比较癌组织与正常组织中miR-10b及其前体和Dicer表达的差异,了解乳腺癌与其表达的关系.方法 通过SYBR Green实时荧光定量PCR方法检测乳腺癌组织和正常乳腺组织中miR-10b及其前体的含量. 结果 (1)miR-10b和Dicer在乳腺癌组织中的表达均显著低于癌旁正常组织中的表达(P<0.05);(2)正常组织中的miR-10b的表达与C-erbB-2成负相关相关 (rs=-0.42 ,P<0.05),正常组织中的Dicer与C-erbB-2的表达与成正相关 ( rs=0.60,P<0.05).结论 miR-10b和Dicer在乳腺癌组织中处于低表达状态,可能在乳腺癌的发生发展中起到抑制作用. |

[Abstract]Objective To study the expression characteristics of miR-10b and its precursor and Dicer in breast cancer.Method We selected 35 patients with breast cancer and collected their cancer tissues and adjacent normal tissues,then used TRIzol to extract total RNA and real-time quantitative PCR to detect the relative gene expression levels. Results The relative expression levels of miR-10b and Dicer in cancer tissue were significantly lower than in adjacent normal tissue(P<0.05). There was a significant correlation between the expression of Dicer and C-erbB-2 (r=0.60,P<0.05)and a significant negative correlation between the expression of miR-10b gene and C-erbB-2(rs=-0.42 ,P<0.05). Conclusion The expression levels of miR-10b and Dicer are low in breast cancer tissues, and may contribute to the inhibition of the development of breast cancer. |
